Have you tried a different cable?

Then I'd suspect that just leaves the possibility of port failure, or corruption of the Android OS. Does plugging in and unplugging the tablet result in  the USB device sound on the PC?

As a workaround, if the files you want to transfer off are in the tablet storage, can you copy them across to a micro SD card and read that card on another device?
