Try these.

To delete corrupted AppLocker files

Right-click at Start and run Command Prompt as Admin
    Type command:
    ren %windir%\System32\AppLocker\Plugin*.* *.bak
    Restart the compuyer

To start the Application Identity Service.

Right-click at Start and click RUN
type SERVICES.MSC and press the enter key
right click on APPLICATION IDENTITY and select START

Restarting is not nessesery for that.

If they dont work, run the system file checker:

Instructions here.(Use  Option 3,To Run SFC /SCANNOW Command in Windows 10)

If the file checker finds problems it cant fix   you can run the DISM Repair Tool.

Instructions here:(Use option 1,To Run "DISM /Cleanup-Image" Tool in Command Prompt)

I gather that it did work at some time. I'm guessing that it failed right after an Update. Do a System Restore at a point just before the crises. If that doesn't fix it, you're in for a reinstall.

You must stay on top of Windows Updates, they are still very sketchy. Win 10 is still very fragile, and subject to the slightest error.
