fat32 and ntfs partition, will it work

I am trying to back up my ps3 to do that I need a external hd with fat32, But I also have data on the hdd.

first off I know to back up anything before formating ok.

my question is can I make 2 partitions, one ntfs and one fat32?

Yes, you may create a mixture of different partition types on a drive. Windows will only offer the FAT32 option on a partition less than 32GB in size.
