oops, now i did it

While out of town my (infected?) email/computer sent something to everyone in my contacts. Several people replied - hacked. One person replied with a link to combofix at the bleeping computer website. I tried to run it and found after I began that I should not have done so without first being requested to do so. Now my computer is spazzing out with a combofix window blinking non stop.
Would someone please help me?

it was opening and closing a great many blue screens every minute. I was able to restore to an earlier setpoint. I am now running malwarebytes as well as the microsoft security essentials scan. When completed I would ask for assistance with the infection.
