External Hard Disk Problem

I've got a 640 Transcend External HD, and I have got a problem, opening one of my folders. It was working fine just a couple of hours ago, and not I can't access it. I have tried chkdsk but nothing works. Now the folder has disappeared and I have no backup. Is there any way I can restore it back?

Does your problem happen on one folder or entire Hardrive?

I have just remembered that two days ago, I made a copy on my desktop and deleted it from the recycle bin. Is there anyway that I can recover it?
