Major Virus issues

I am working on my future father inlaws pc due to major virus infestation. He is running Windows Home edition SP2. He has Mcafee b/c he got it for free. I have tried Malewarebytes (both in and out off safemode) but the virus would not let it run. I have also tried combofix to no avail. He called Mcafee and all they did was send him an email to a free scanner. I believe I have noticed AntiVirus suite 2010 and AntiVirus8 and there are probably more. they will not let anything run. Any suggestions
