Defense Security Virus. I can't remove it.

I noticed I got the Defense Security virus today and I've been working for many hours trying to remove it. I've been able to access task manager and remove a few things, and my antispyware run. My computer is running slow now, and under my roaming folder I found a file for it and when I tried to delete it it said access was denied. I deleted some files from inside of the folder, but there's one called defcnt it won't let me delete. How can I remove this?

Mcafee and Superantispyware I've had run scans on the folder it won't let me delete, and neither catch it. Whenever I try to manually delete the folder it says destination folder access denied. I downloaded malwarebytes but it's not opening to do a scan.
