In System Preferences > Startup Disc, is the flash drive listed when you connect it?
It might help if you posted which Mac you have and which OS X is installed on it in order to get better help from someone.

Choose it and hit the "Restart" button, the Mac should boot from the flash drive.

Connect the flash drive.
Go to System Preferences > System > Startup Disk.
Select the flash drive drive then hit the Restart button.
You did say the flash drive shows up as a start up disk in System Preferences right?
