NEED ASSISTANCE ASAP<----VIRUS TOOK OVER

Ok, Ive gone through the steps provided at http://www.bleepingcomputer.com/combofix/how-to-use-combofix

However, the Trojan has a mind of its own and wont let anything spyware removal or antivirus programs to run. It erases them. I ran the comp in safe mode but cant do anything to fix my comp. When I try to run it in regular mode I can log into my account but all it does is go to a black screen and pops up error dialogs saying that "blah blah" has crashed or is no longer working. Either I close it or say to look up a fix for it later. Obviously that wont work. It just sits at that screen and cycles through all the things that wont work.

My files are all on an external harddrive so I'm not losing anything, but I still need my computer. What do I do?
