Can't get rid of 'Total Security' (virus)

I keep getting pop-ups telling me my computer is infected and to buy Total Security. I have never indicated I wanted it, but it is on my computer now and I can't get rid of it. It has it's own desktop wallpaper indicating my computer is infected. It won't let me access Windows Task Manager, Control Panel, my anti-virus program, or the Internet. I have been advised to use your Combo Fix but information provided about this states I should do this under direction. I have downloaded ComboFix to a USB stick - I know to start my computer in safe mode and run this. What else should I know?
