Can I remove a USB which is used as RAM in computer?

Click Start, navigate to Computer on the right (what used to be known as My Computer), click.

Right-click the removable drive being used for ReadyBoost, click Properties, click Advanced tab and select "Do not use this device".

Remove USB.
