We have a legacy code that works on DOS and it communicates on a RS-232 serial Port,
Please let us know if there is any procedure to determine the protocol(Kermit/X/Y/Z modem)
it is supporting on the serial port.

Little confused as to what you are asking. You wont be able to use ftp over a serial port if that is what you want to know
